Preparation of Samples for Mycoplasma Detection in Cell Lines
The customer provides 1-2 ml of the culture medium. In order to ensure that a reliable result is obtained, the following recommendations must be observed when preparing the medium sample: - Upon thawing the cell stock from liquid nitrogen, cultivate the cells under standard culture conditions for at least 14 days.
- Cultivate the cells in a volume of culture medium recommended for the given culture dish (e.g., 5 ml of the medium for the culture flask with the area content of 25 cm2); do not exceed maximum recommended volume.
- Cell confluence at the time of the medium collection must be at least 80%.
- Collection of the medium sample must be performed not before 2 days after the last medium replacement.
- Centrifuge 1-2 ml of the collected culture medium (1500 rpm; 5 minutes; at room temperature) and using a pipette transfer the medium to a new test-tube.
- Keep the medium for 14 days at the maximum, at -70 °C.
- Temperature of the samples must not exceed 25 °C during their transport from the customer while the transportation time must not exceed 48 hours.
