Plasmid Amplification and Isolation

Amplification and isolation of plasmids is provided as part of our molecular services. These services include transformation of the supplied plasmid into bacteria Escherichia coli, its amplification in the bacterial culture and subsequent isolation.

Yield of the plasmid isolation depends on many factors, especially on the sequence for the starting point of DNA replication (the so called ORI), plasmid size and insert properties (for example, the inserted shRNA/miRNA sequence may decrease yield of the plasmid). In order to achieve maximum yield, it is advisable to supply information on the plasmid classification in the "high copy" or "low/very low copy" group based on recommendations in the table below, together with the plasmid. We cannot bear responsibility for the total yield upon improper classification of the plasmid. Upon receipt of more detailed information about the plasmid (length, name of the original vector and map of the plasmid), we shall classify the plasmid in the proper group ourselves.

Terms of delivery

Plasmid isolate is dispatched by post on ice within 15 working days upon receipt of your purchase order.

The plasmid is dispatched as solution in the TE buffer. Furthermore, the customer obtains information on its concentration and purity.

Requirements for the materials

The plasmid can be provided as either a solution or a dry spot on the filter paper.

  • Minimum plasmid quantity in the solution: 100 ng
  • Minimum plasmid quantity on the filter paper: 1 µg
  • Minimum purity (A260/A280) 1.8 at the minimum

Furthermore, we need that the customer specifies the antibiotic resistance gene and classification of the plasmid in the "high copy" or "low/very low copy" group.
