Oligonucleotide PurificationGENERI BIOTECH provides 3 ways of oligo purification: - Standard purification - good-quality synthesized oligonucleotides approx. 40 - 50 bases long show such a low proportion of chains not completely synthesized that their purification is unnecessary and only leads to reduced yield. In indicated cases, desalting is performed on the column filled with the medium Sephadex G-25. The resulting product is cleared of chemical impurities (excessive amount of salts, protective groups).
- OPC purification (Oligonucleotide Purification Cartridge) - oligonucleotides containing the trityl group are captured on the lipophilic sorbent of the OPC column (shorter chains not completely synthesized contain no trityl groups). The resulting product is cleared of chemical impurities (excessive amount of salts, protective groups) and shorter chains not completely synthesized. OPC purification shows the most favourable quality-to-price ratio. Purity of the resulting product is comparable to HPLC purification and is sufficient for all common applications.
- HPLC purification (trityl-off) - oligonucleotide is separated from all impurities and improper length sequences. However, this mode of purification is very demanding and amounts to significant losses.
