Sequencing

DNA sequencing is a fundamental, irreplaceable method in molecular biology. Precisely for this reason, it is a pivotal technique at GENERI BIOTECH that is used on the routine basis for our own research. Our experience and other applications of DNA sequencing are offered as part of our molecular services.

What is provided:

• Routine sequencing of plasmids and PCR products (the so called standard sequencing)
• Identification of bacteria based on sequencing (16S/ITS segments)
• Implementation of more complex sequencing projects

Two automatic sequenators are available in our laboratory (ABI PRISM® 3100-Avant Genetic Analyzer, Applied Biosystems) that provide reduced risk of service interruption. Sequenators operate on the principle of capillary electrophoresis with fluorescent detection. "Dye-terminator" type of sequencing reaction is performed as standard where unlabelled primer and four fluorescent labelled dideoxynucleotide terminators are used.

Evaluation of data obtained by means of this method is performed automatically by the sequenator and checked manually by a qualified employee. A sequenogram is the result - graphic output in the form of coloured peaks (scf files). The sequence is well readable (i.e. accuracy > 99%) approximately starting from 20-30 bases beyond the primer up to 600 bases.
