Standard Sequencing

Standard sequencing includes sequencing of DNA cloned into the plasmid vector or PCR product sequencing. Plasmids are sequenced by universal primers (stated in the list) as well as primers provided by the customer. Amplification primers provided by the customer are used for PCR product sequencing. In the case of specific requirements, we design and synthesize the primers ourselves.

Results are sent via e-mail or by post on the CD within 5 working days upon receipt of the purchase order.

The sequences are provided to the customer in the form of scf files with a clearly arranged summary of the results in a table (the so called sequencing key). Scf files can be viewed and modified using applications freely available on the Internet, for example:

How to place an order

Orders of standard sequencing can be submitted via e-mail to the address ; with the word SEQ stated in the subject of the e-mail. You can also print the completed purchase order form and send it by post together with the samples (and possibly with the primers) to our address.

Requirements for the material

Samples and primers should be prepared to 1,5-2 ml tubes in deoinized water or 10mM Tris buffer, not TE buffer.

  • Plasmid DNA sequencing:

- Purity (A260/280) 1.8 at the minimum
- Minimum DNA concentration 100 ng/µl
- Minimum sample volume of 20 µl

Concentration and purity of the plasmid can be determined by means of spectrophotometry.

  • PCR product sequencing:

1) Purified samples

- Minimum DNA concentration 50 ng/µl
- Minimum sample volume of 20 µl

2) Unpurified samples

- Minimum DNA concentration 100 ng/µl
- Minimum sample volume of 30 µl

Quality and quantity of the PCR product should be verified by the customer by means of electrophoresis. Please, insert the photograph and data on electrophoresis in to the order form. You can also obtain information on input quality of the PCR product from requirements for the PCR product quality.

  • Sequencing primers provided by the customer should meet these criteria:

- Minimum primer concentration 10 pmol/µl
- Minimum primer volume of 20 µl (enough amount for 5 sequencing reaction)
- Tm ≥ 50 °C (optimal Tm is 50-60 °C)
- Length 18-25 base pairs
- GC content 30-70%
- Primers should not form any strong dimers or hairpin loops
