Standard oligonucleotides reliably cover routine laboratory needs. However, more complex experimental designs—such as the preparation of long control templates, specific probes, or advanced genetic manipulations—often require sequences exceeding one hundred bases. To address these specific research applications, GENERI BIOTECH has expanded its portfolio with a new offering: ultramers.
Standard Synthesis Parameters
Ultramers are a targeted solution for synthesizing long oligonucleotide sequences ranging from 100 to 130 bases. By default, they are synthesized at a 40 nmol scale using standard purification. This specific setup ensures an optimal balance between the final yield and the overall quality of the synthesis.
Quality Control
At GENERI BIOTECH, we routinely use MALDI-TOF mass spectrometry for oligonucleotide quality control (QC). However, the high molecular weight of ultramers pushes the analytical limits of our instrumentation, exceeding the standard detection thresholds of the MALDI-TOF method. Consequently, we cannot provide a fully guaranteed QC readout for sequences of 101 bases and longer.
Flexibility for Custom Designs
Although standard synthesis covers lengths up to 130 bases, real-world experiments frequently demand custom sequence modifications.
Common requirements include:
- Introduction of functional modifications at the 3′ and 5′ ends
- Specific internal sequence modifications
- Alternative synthesis scales or advanced purification methods
- Exceptionally long sequences exceeding 130 bases (131-mers and longer)
These non-standard ultramer variants can often be manufactured following an individual feasibility assessment. In such cases, we arrange a direct technical consultation with our production department.
