Kit for isolation of DNA from microorganisms

Kit for isolation of DNA from microorganisms

The DNA extraction kit enables isolation of microbial DNA from pellet of bacterial cells, yeasts and fungi. It is possible to use it for genomic DNA isolation from different types of tissues of both human and animal origin. High DNA yields, reproducible results and simple working method are guaranteed. All sample preparation buffers are included.

Principle of the isolation kit

  • Cell lysis is reached by mechanical disruption using beating beads and followed by incubation of the particular sample with Proteinase K and solutions with a higher content of chaotropic ions.
  • DNA isolation is performed on the silica membrane of the column.
  • Specific conditions appropriate for binding DNA onto a membrane are achieved by ethanol added to lysate.
  • Consequent treatment by two types of buffer results in removing impurities in the first step and eluting pure genomic DNA in the next step (ratio A260/A280 1.70 to 1.90).

Dual-labeled hydrolysis probes Dual-labeled hydrolysis probes

The technology of dual-labeled hydrolysis probes (also called TaqMan® real-time PCR probes) exploits the 5 ́-3 ́ exonuclease activity of Taq DNA polymerase to cleave the probe. TaqMan real-time PCR probes are labeled with a fluorescent dye (reporter) at the 5´ end and with a quencher at the 3´ end.

Background Probiotics are defined as live microorganisms which, when administered in adequate amounts, confer a health benefit on the host. For instance, probiotics can be used in the treatment and prevention of infections and chronic inflammatory disorders of the gastrointestinal tract. Due to their health attributes, probiotics generate considerable interest in the area of functional
