Plasmid verification

This service is recommended when it is necessary to verify whether a correct sequence was cloned in, whether plasmids were not misidentified (mistaken by accident), or to verify that there are no errors in the plasmid sequence.

Description of the service

The following methods are most commonly used to verify accuracy of a plasmid:

1) Restriction analysis enabling the recognition of more extensive deletions (typically if a target sequence was not cloned in) or an accidental interchange of plasmids (typically if plasmids do not originate from an original source).

2) Plasmid conformation determination suitable for quality check of a native plasmid. When native, a plasmid should be present prevailingly in supercoiled conformation. In order to verify the conformation, plasmid analysis on gel is used and a percentage of the supercoiled form is determined.

Plasmid verification can also be performed under GMP or GLP regulations.

Output of the test

We provide the customer with a reporting protocol. If the customer determines acceptability criteria, it is possible to issue a report stating that the plasmid complies with the determined parameters.
