Phosphorothioates contain the exchange of one oxygen in the phosphate bond with sulfur. By exchanging oxygen for sulfur, the resistance of the oligonucleotide to nucleases increases.


pure phosphorothioates: substitution is made in all phosphate linkages in the chain
mixed phosphorothioates: substitution is only made on selected phosphate bonds.

Phosphorothioates are used for antisense applications where a 10 μmol scale is usually selected.
